What is GlyNAC?
GlyNAC is not a single substance but rather a combination of glycine and N-acetylcysteine (NAC). Both are precursors to the body’s own antioxidant glutathione — the most important intracellular protective molecule.
The problem: Glutathione production declines sharply with age. NAC alone supports the cysteine component, but glycine — the second amino acid — is often deficient. GlyNAC supplies both building blocks together and is therefore more effective than supplementing each individually.

Is GlyNAC safe?
Both individual substances are well-characterized and have been used for decades. NAC: at very high doses (>3g daily) may cause gastrointestinal side effects. Glycine: very safe. Use NAC cautiously in asthma.
How does GlyNAC work?
Glutathione (GSH) is synthesized in two enzymatic steps: γ-glutamylcysteine synthetase combines glutamate and cysteine, then glutathione synthetase adds glycine. Age-related glutathione decline occurs due to decreased activity of these enzymes and reduced amino acid availability. NAC provides bioavailable cysteine (not found in high quantities in food), while glycine is also often limiting in older adults’ diets. Together, they restore the substrate for glutathione synthesis.
Evidence timeline
2021 Kumar et al. landmark study (n=24, 24 weeks): Older adults (mean age 73) received GlyNAC (2.4g glycine + 0.8g NAC daily) or placebo. Results:
- Plasma glutathione increased to levels seen in young healthy adults (~900 vs 560 µmol/L)
- Intracellular glutathione improved in peripheral blood mononuclear cells
- Mitochondrial hydrogen peroxide production decreased (less oxidative stress)
- Inflammatory markers (TNFα, IL-6) declined
- Muscle strength (grip strength) improved
- Cognition scores improved on standard testing
- Physical function (gait speed, leg strength) improved
2022 Kumar follow-up (energy metabolism): In the same cohort, GlyNAC improved fatty acid oxidation in mitochondria — a key marker of metabolic health. Body composition shifted toward less fat mass.
2022 Murine lifespan study: Mice given GlyNAC (dosed by body weight equivalent) lived 24% longer than controls, with reduced markers of aging, improved metabolic markers, and preserved immune function.
Why this matters for aging
Glutathione is called the “master antioxidant” because it exists in every cell and neutralizes free radicals, supports detoxification, and stabilizes mitochondrial function. Declining glutathione is associated with nearly every age-related disease: neurodegeneration, cardiovascular disease, muscle wasting, metabolic dysfunction, and cancer. NAC at doses >3g daily can inhibit copper absorption and has rare reports of rash or other side effects. Long-term high-dose NAC safety in aging populations has not been formally studied in multi-year trials.
Practical considerations
Dosing: The studies used 2.4g glycine + 0.8g NAC daily (total 3.2g). This is achievable by buying glycine and NAC separately — significantly cheaper than branded “GlyNAC” products, though those also exist.
Timing: NAC is better absorbed on an empty stomach; glycine can be taken anytime.
Who might benefit most: Older adults (70+) showing signs of muscle weakness, cognitive decline, or metabolic dysfunction. Less clear for younger, healthy individuals.
Who should be cautious: Anyone with cystic fibrosis, severe kidney disease, or asthma should discuss NAC with their doctor. NAC can theoretically affect blood clotting medications (rare but documented).
The verdict for longevity
GlyNAC stands out because it bridges the gap between mechanistic plausibility and actual human data. We understand why glutathione matters for aging. We see that it declines with age. And we now have evidence that restoring it improves markers across multiple aging dimensions — not just one biomarker, but strength, cognition, mitochondrial function, and inflammation simultaneously.
Is it a longevity game-changer? Too early to say. But for older adults seeking affordable, evidence-based strategies to slow aging, GlyNAC deserves serious consideration — especially as part of a broader healthy aging toolkit (exercise, sleep, nutrition, stress management).
