What Is L-Theanine?
L-theanine is a non-proteinogenic amino acid found naturally in tea leaves (Camellia sinensis). Green tea typically contains 20–40 mg theanine per 200 ml cup; matcha provides considerably more (~50–100 mg per serving). Outside of tea, L-theanine is rarely found in foods—one exception being the mushroom Xerocomus badius.
As a supplement, L-theanine is available in capsules or powder, typically standardized to 100–200 mg per dose. The compound has been studied most extensively in Japan in the context of green tea consumption and ranks among the best-selling nootropics worldwide.
Mechanism: How Does L-Theanine Work?
Pharmacological and neuroscience studies have identified several mechanisms of action:
Alpha-wave activation: EEG studies consistently show that as little as 50 mg L-theanine significantly increases alpha-wave power in the brain. Alpha waves (8–12 Hz) are characteristic of relaxed, focused wakefulness—a state often described as “calm focus.”
Glutamate antagonism: Theanine is structurally similar to the excitatory neurotransmitter glutamate and competitively binds to NMDA and AMPA receptors without fully activating them. This can dampen excessive neuronal excitation.
GABAergic modulation: Evidence suggests modulation of inhibitory GABAergic signaling, which may contribute to the relaxing effect without classical sedation.
Caffeine interaction: Theanine and caffeine interact complementarily at the adenosinergic system. Theanine can attenuate some undesirable effects of caffeine (anxiety, elevated heart rate in sensitive individuals) without suppressing wakefulness.
What Does the Evidence Actually Show?
Reasonably Consistent Findings
Alpha waves on EEG: Multiple studies confirm measurable increases in alpha-wave power at 50–200 mg L-theanine. This is the most robust biomarker for L-theanine’s effect.
Stress reduction: In small crossover studies, L-theanine (200 mg) reduced physiological stress markers such as heart rate and salivary IgA levels during mentally stressful tasks (Kimura et al. 2007, n=12).
Theanine + caffeine — synergistic effect: Multiple RCTs show that the combination of L-theanine and caffeine (~50 mg theanine + 75 mg caffeine) consistently outperforms either substance alone on attention tasks. This is the best-supported use case (Haskell et al. 2008, n=27; Dodd et al. 2015, n=24).
Sleep quality: A randomized study in boys with ADHD showed improved objective sleep quality (actigraphy) at 400 mg/day without sedative side effects (Higashiyama et al. 2011, n=98).
Important Limitations
- Most studies are very small (n<30) and typically single-session acute crossover designs. Long-term effects over weeks or months are hardly studied.
- L-theanine alone (without caffeine) often shows no significant cognitive improvements vs. placebo in healthy, well-rested adults.
- Subjective stress scales are vulnerable to placebo effects; separating true pharmacological effect from expectation is difficult.
- Effect sizes are moderate and clinically unspectacular: reaction time gains are typically in the 10–30 ms range—measurable, but barely noticeable in daily life.
- Reproducibility across independent labs is less consistent than for caffeine or creatine.
Bottom line: The combination with caffeine is far better supported than L-theanine as a standalone supplement. Without a caffeine context, expectations should be modest.
Tea vs. Supplement
A typical 200 mg L-theanine supplement provides the theanine equivalent of approximately 5–10 cups of green tea—which would also deliver substantial caffeine (~150–300 mg). Isolated supplementation makes sense when you want to deliberately control the caffeine-to-theanine ratio.
The well-studied caffeine:theanine ratio is 1:2 (e.g., 100 mg caffeine + 200 mg theanine). Matcha naturally provides a similar ratio (~1:2 to 1:3).

Safety
L-theanine is considered likely safe at standard doses. In Japan, it has been approved as a food additive (“FOSHU”) for decades. Human studies have used up to 400 mg/day without adverse effects. Data for higher doses or long-term use over months is limited.
Caution with concurrent blood pressure medications (mildly additive blood pressure–lowering effects possible) and in individuals sensitive to glutamate modulators.
Summary
L-theanine is one of the more reasonable nootropics: no miracle effects, but measurable—if moderate—evidence for relaxation and synergistic benefits with caffeine. As a 1:2 combination with caffeine, it is well-studied and a practical option for many people. As a solo supplement without caffeine, the evidence is weaker. Hype claims (deep sleep, cognitive enhancement, anti-aging) are not supported by the existing literature.
